摘要
目的:分析测定23例健康人CD3+CD4+T淋巴细胞计数与相对时间之间的关系。方法:对采集的23例全血标本以FITC-CD4/CD8/CD3抗体标记,通过流式细胞仪分别在采血后0 h,24 h,48 h,72 h,96 h进行CD3+CD4+T淋巴细胞计数。应用SPSS 13.0统计软件进行重复测量资料的方差分析,再应用General Linear Model(GLM)对后四次与第一次检测结果之间的差异做方差分析,评估CD3+CD4+T淋巴细胞计数的最佳时间及后4次检测结果与第一次之间的差异。结果:球形检验的结果判断重复测量数据之间存在相关性(P<0.05),在Greenhouse-Geisser校正下应用GLM模型分析后四次与第一次计数之间的差异均有统计学意义(P<0.05),计算后四次与第一次计数之间的下降率,分别为1.0%、1.8%、21.5%、31.9%。结论:应用FITC-CD4/CD8/CD3抗体标记全血标本,实验室检测CD3+CD4+T淋巴细胞计数在48 h内完成,结果较能反映受试者的真实情况。
Objective:To determine the relationship between CD3 + CD4 + T lymphocyte count of 23 healthy persons and the relative time.Methods: Mark the collected whole blood specimens of 23 healthy persons by FITC-CD4/CD8/CD3 antibody.Then take count of CD3 + CD4 + T lymphocyte by flow cytometry,respectively after blood sampled 0 hours,24 hours,48 hours,72 hours,96 hours.Analysis the test datum by the repeated measurement data analysis of variance through statistical software SPSS 13.0.Then test the variance between the 1st and other four times by the GLM procedure to assess the best time for the detection of CD3 + CD4 + T lymphocyte count.Results: It results from Mauchly′s Sphericity Tests that correlation exists in repeated measurement datum(P0.05).There is statistically significant between the1st and all the other four times,which depends on the results of the GLM procedure adjusted by Greenhouse-Geisser(P0.05).The droop rate between the posterior four times and the 1st are respectively 1.0%、1.8%、21.5%、31.9%.Conclusion: It is more reliable that the result comes from the flow cytometry which completed in 48 hours for CD3+CD4+T lymphocyte counts.
