Optimization of macroporous resin purification of crude ferulic acid by response surface methodology

摘要 利用响应面法对大孔树脂纯化阿魏酸粗提液工艺进行优化,在单因素试验基础上,根据中心组合设计原理,采用三因素三水平响应面分析法,依据回归分析确定最优纯化工艺条件,得出最优工艺为:吸附pH值6.2、洗脱剂乙醇浓度58.22%、洗脱剂pH值7.1;在该工艺条件下,阿魏酸回收率达97.036%,且经薄层层析法验证,纯度大大提高,可达75.65%。 Based on the single factor experiment,the response surface method with three factors at three levels is adopted according to the principle of central compares design in order to optimize the purification conditions for the crude ferulic acid from wheat bran.The factors of optimum conditions for the purification were determined by regression analysis.The results showed adsorption pH value of 6.2,ethanol concentration of 58.22%,and elution agents pH value of 7.1.In these conditions,the ratio of ferulic acid recovery is 97.036%.The purity is greatly improved through the TLC analysis the ferulic acid purification rate is 75.65%.
