Dnmt1、Caveolin-1在结直肠癌中的表达及其临床意义

Clinical Significance and Expression of Dnmt1 and Caveolin-1 Protein in Colorectal Carcinomas

目的探讨Dnmt1和Caveolin-1在结直肠癌中的表达及其临床病理意义。方法应用免疫组化PV-9000二步法检测84例结直肠癌组织及20例癌旁正常组织中Dnmt1、Caveolin-1蛋白的表达情况,分析二者与其临床病理特征的关系。结果 Dnmt1蛋白在结直肠癌中的阳性表达率(72.62%)显著高于癌旁正常组织(25.00%)(P<0.05);Caveolin-1蛋白在结直肠癌中的阳性表达率(32.14%)显著低于癌旁正常组织表达率(75.00%)(P<0.05);二者表达与肿瘤的组织学分化程度、Dukes分期、浸润深度、淋巴结转移等组间差异性有显著性意义(P<0.05);Dnmt1和Caveolin-1蛋白的表达呈负相关关系(r=-0.304,P=0.000)。结论 Dnmt1和Caveolin-1的表达与结直肠癌的发生、发展密切相关。

Objective To investigate the expressions and their clinicopathological significance of Dnmtl and Caveolin - 1 proteins in coloreetal cancer. Methods The expressions of Dnmtl and Caveolin - 1 in 84 cases of colorectal carcinomas and 20 cases of adjacent normal mucosa were detected by PV - 9000 two - step immunohistochemical method. The relationship between their expressions and the clinicopathologic features of colorectal cancers were analyzed. Results The positive rate of Dnmtl in colorectal carcinomas （72. 62% ） was significantly higher than that in normal tissues （25.00%） （P 〈0. 05）. The positive rate of Caveolin - 1 in colorectal carcinomas （32. 14% ） was significantly lower than that in noral control tissues （75.00%） （P 〈 0. 05）. Both the expressions of Dnmtl and Caveolin - 1 were associated with histologi- cal differentiation, serous membrane invasion, lymph node metastasis and Dukes stage of colorectal carcinoma （P 〈 0. 05）. The expression of Dnmtl was negatively correlated with that of Caveolin - 1 （ r = - 0. 304, P = 0. 000）. Conclusions The expressions of Dnmtl and Caveolin - 1 are closely related to carcinogenesis and the development of colorectal carcinonlas.
