摘要
目的观察疏肝和胃降逆颗粒对反流性食管炎大鼠胃和食管pH值、食管黏膜增生及PCNA表达的影响。方法 50只SD大鼠随机分为正常组、假手术组、模型组、疏肝和胃降逆颗粒组和奥美拉唑组,每组10只。用幽门半结扎加食管下括约肌切开法建立反流性食管炎大鼠模型,造模1周后给予药物干预,共28 d。治疗结束后处死动物,进行食管下段及胃pH值检测,取食管下段组织,分别进行常规HE染色观察黏膜增生和采用QRT-PCR技术法检测PCNA的表达。结果模型组食管下段和胃的pH值较正常组明显下降(P<0.05);模型组食管下段黏膜增生和PCNA的表达较正常组和假手术组均明显升高(P<0.05);疏肝和胃降逆颗粒组和奥美拉唑组食管下段和胃pH值均较模型组明显升高(P<0.05);疏肝和胃降逆颗粒组和奥美拉唑组食管下段黏膜增生和PCNA的表达较模型组均明显降低(P<0.05)。疏肝和胃降逆颗粒组和奥美拉唑组比较,上述各指标差异没有统计学意义(P(0.05)。结论疏肝和胃降逆颗粒对反流性食管炎大鼠胃和食管pH值、黏膜增生和PCNA的表达均有明显的抑制作用。
Objective To study the influence of SGHWJN particles on gastroesophageal PH value,esophageal mucosal hyperplasia and expression of PCNA in esophageal tissue of rats with reflux esophagitis.Methods Fifty SD rats were randomly divided into 5 groups,namely control group,sham-operated group,model group,SGHWJN particles group and PPI group.Reflux esophagitis was induced by adopting partial pyloric ligation plus cardiomyotomy.One week later,the rats were orally administered twice daily for 28 days.Pathological changes of esophagus mucous membrane were evaluated by HE staining.Gastroesophageal pH was determined by pH Meter and expression of PCNA in esophageal tissues was examined by real-time fluorescent quantitative reverse transcription polymerase chain reaction(RT-FQ-PCR) with SYBR Green.Results Operated gastroesophageal PH value of model group was significantly lower than that of control group and sham-operated group(P0.05);esophageal mucosal hyperplasia and expression of PCNA in esophageal tissues were significantly higher,compared with normal group and sham group(P0.05);gastroescophaspeal pH value in SGHWJN particle group and PPI group of model group was significantly higher than that of model group(P0.05);esophageal mucosal hyperplasia and expression of PCNA in esophageal tissues of SGHWJN particles group were decreased significantly as compare to control group and PPI group(P0.05).No statistically significant difference could be observed between the normal group and sham-operated group or between SGHWJN particle group and PPI group(P 0.05).Conclusion SGHWJN particles can effectively inhibit gastroesophageal PH value,esophageal mucosal hyperplasia and expression of PCNA in esophageal tissues in rats with reflux esophagitis.
