摘要
为探索出北冬虫夏草液体菌种制备的适宜碳源、氮源及配方,以葡萄糖、果糖、蔗糖、麦芽糖、淀粉及硝酸钾、硝酸铵、尿素、蛋白胨、酵母膏和牛肉膏等,以及马铃薯、小麦、蚕蛹粉、蛋清、鱼粉和黄豆粉等单一或复合碳源、氮源,组合不同配方及不同碳氮比,以单因子对照方式,比较观察菌种菌丝及其栽培发菌、抗感染及出草情况。结果表明:单一碳源中,葡萄糖最优;复合碳源居中,葡萄糖+淀粉最佳;常见碳源原料中以土豆最优;常见氮源原料中以有机氮源优于无机氮源,蚕蛹粉>蛋清液>鱼粉>黄豆>小麦;最佳C/N为5∶1。蚕蛹、蛋清、土豆等常见原料可作为菌种制备的优选原料及配方为马铃薯(去皮)200 g+葡萄糖10 g+蚕蛹粉30 g(煮汁)+硫酸镁2 g+磷酸二氢钾2 g+VB110 mg+水1 000 mL+pH 6.5,马铃薯(去皮)200 g+葡萄糖10 g+蛋清液30 g+硫酸镁2 g+磷酸二氢钾2 g+VB110 mg+水1 000 mL+pH 6.5。
The single-factor tests were conducted to study the effect of different carbon source,nitrogen source and formula on mycelial growth and infection resistance of liquid Cordycps militarisc strain for probing the suitable carbon source,nitrogen source and formula.The results showed that glucose,glucose + starch and potato was the best in sole carbon source,compound carbon source and common carbon source respectively,the organic nitrogen source was better than inorganic nitrogen source in common nitrogen source,the optimum C-N ratio was 5∶1,and the optimum media for good quality of liquid Cordycps militarisc strain were 200g peeled potato+10g glucose+30g silkworm chrysalis powder(Boiled juice)+2g MgSO4+2g KH2PO4+10mg VB1+1000mL water+pH 6.5 and 200g peeled potato+10g glucose+30g egg white liquid+2g MgSO4+2g KH2PO4+10mg VB1+1000mL water+pH 6.5.
