大蒜休眠芽愈伤诱导和植株再生 被引量:1

DEVELOPMENT OF A PLANT REGENERATION SYSTEM FROM DORMANT BUD-DERIVED CALLUSES OF GARLIC (ALLIUM SATIVUM L.)

摘要 本研究以优良品系‘金乡大蒜’休眠芽为外植体,成功地建立了再生体系。基本培养基为MS,在添加了2,4-D 2.mg/L的培养基上最适于休眠芽愈伤诱导,愈伤诱导率为99.5%,黄色颗粒愈伤诱导率为100%,愈伤最佳继代培养基为MS+24,-D 2.0 mg/L;在最佳分化培养基MS+6-BA3.0 mg/L上,黄色愈伤分化率为98.6%,芽最佳生长培养基为MS+6-BA1.0 mg/L+KT1.0 mg/L+GA0.2 mg/L+NAA0.1 mg/L,且适于芽生长的最佳温度为20℃;试管苗最佳生根培养基为MS+NAA0.2 mg/L,生根率达到92.4%;生根的试管苗移栽到装有田园土的土盆中,其存活率达到82%。本试验为今后开展大蒜遗传转化和体系胞突变体筛选工作奠定了良好的实验基础。 Plant regeneration systems have been successfully established with dormant bud of Jin garlic No. 3' as explant. The basal culture medium was MS, The optimum medium of callus induction was MS medium with 2, 4 -D2.0 rag/L, the rate of callus induction was 99.5%. The rate of yellow granule callus induction could be 100%, the optimum medium of callus subculture was MS medium with2,4 - D 2.0 mg/L. The rate of yellow granule callus differentiation could be 98.6% on optimum medium with 6 - BA 3.0 mg/L. The optimum me- dium of shoot growth was MS medium plus 6 - BA1.0 rag/L, KT1.0 mg/L, GA0.2 mg/L and 0.1NAA mg/L. The optimum temperature of shoot growth was20℃. The rooting rate was 92.4% on optimum rooting medium with NAA0.2 mg/L. The survival rate of plant transplanting was 92.4% after transferred to pots with garden soil. This experimentation may provide basic reference for screening of somaclonal variants and genetic transformation of garlic.
