摘要
目的研究bcl-2反义寡核苷酸在抑制HL-60细胞增殖和下调细胞内bcl-2蛋白表达水平的作用。方法将人工合成的bcl-2反义寡核苷酸片段与HL-60细胞共培养,在作用的第0天、1天、3天、5天通过细胞计数和锥虫蓝染色法检测细胞的生长、存活情况,同时通过免疫组化法(APAAP法)检测细胞内bcl-2蛋白表达水平的变化情况。结果bcl-2正、反义寡核苷酸均可抑制HL-60细胞增殖,但bcl-2反义寡核苷酸的作用更为显著。bcl-2反义寡核苷酸可下调HL-60细胞内bcl-2的蛋白表达水平,其作用与其浓度和时间呈正相关,而bcl-2正义寡核苷酸及对照无此作用。结论bcl-2反义寡核苷酸可以有效地抑制HL-60细胞的增殖和降低细胞内bcl-2的蛋白表达水平。
Objective To investigate the effect of bcl-2 antisense oligodeoxynucleotides on inhibiting the growth of HL-60 cells and reducing the level of bcl-2 protein expression. Methods Synthesized bcl-2antisense oligodeoxynucleotides were added to in vitro HL-60 culture system, We used cell counts and trypan blue dye to detect the cell growth condition,and used immunocytochemical staining assay to detect the level of bcl-2 protein expression on day 0, 1, 3, 5. Results bcl-2 sense and antisense oligodeoxynucleotides may inhibit the growth of HL-60 cells, but the effect of bcl-2 antisense oligodeoxynucleotides was more significant. bcl-2 antisense oligodeoxynucleotides may reduce the level of bcl-2 protein expression in HL-60 cells by the dose-dependent manner and time-dependent manner, but there was no change in the sense oligodeoxynucleotides and the control. Conclusion bcl-2 antisense oligodeoxynucleotides inhibited the growth and reduced the level of bcl-2 protein expression in HL-60cells.
