摘要 目的:建立刺五加口服液中刺五加总皂苷含量测定的UV法。方法:采用AB-8型大孔吸附树脂分离刺五加总皂苷[1],利用此类化合物的分子结构皆为三萜皂苷,在齐墩果烷型配基上连有单糖链或双糖链,与香草醛浓硫酸加热反应生成物在540nm处显最大吸收[2]的特性,选用齐墩果酸为对照品,用UV法测定刺五加总皂苷的含量。结果:该方法在20.96μg^125.76μg浓度范围内具有良好的线性关系,R=0.9999;平均回收率为99.64%,RSD=0.56%。结论:该方法准确,可靠,重复性好,可作为刺五加口服液质量控制的方法。 Objective:To establish UV method for determination of the content of the total saponins of Acanthopanax senticosus.Methods:In this article,the total saponins of Acanthopanax senticosus were separated by AB-8 macroporous adsorption resin.Molecular structures of these compounds were all triterpenoid saponins,whose oleanane type ligands were equipped with monodesmosidic or bidesmosidic.The resultants of their heated reactions with vanillin sulfuric acid had maximum adsorption at 540 nm.Considering these characteristics,oleanolic acid was chosen as the reference substance to determine the total saponins of Acanthopanax senticosus by UV method.Results:The standard curve was in good linearity over the range of 20.96μg~125.76μg,R=0.9999 for determination of its related substances.The average recovery rate was 99.64%,RSD=0.56%.Conclusion:The method is accurate,reliable and reproducible and can be used for the quality control of Acanthopanax senticosus oral liquid.
