稳心颗粒对大鼠心室肌细胞L型钙电流的影响 被引量:17

摘要 目的研究步长稳心颗粒对大鼠心室肌细胞L型钙电流((L-type calcium current,ICa-L)动力学特性的影响。方法以酶解法分离获得单个心室肌细胞,采用全细胞膜片钳技术,研究1g/L、5g/L、10g/L稳心颗粒对ICa-L的影响。结果 1g/L、5g/L、10g/L的稳心颗呈浓度依赖性地抑制ICa-L,对其峰电流的的抑制率分别为29.3%±4.8%、45.8%±5.3%、72.6%±4.1%。10g/L的稳心颗粒使ICa-L电流电压曲线上移,激活曲线右移,并延长失活后恢复时间。结论稳心颗粒对大鼠心室肌细胞ICa-L具有浓度依赖性的抑制效应,与其抗心律失常效应有关。 Objectives To study the effects of Wenxin granule on the activation kinetics of L-type calcium current (ICa-L) in single ventricular myocyte. Methods The whole-cell patch-clamp technique was used to investigate the effects of Wenxin granule( 1 g/L, 5 g/L and 10 g/L) on ICa-L in acutely isolated ventricular myocytes of adult rat. Results Wenxin granule particle inhibited the current densiyty of ICa-L, and the decreased rates of the peak ICa-L were 29.3%±4.8%, 45.8%±5.3% and 72.6%±4.1% at 1 g/L,5 g/L and l0 g/L group,respectively.The current-voltage curve was shifted upwards,and the steady-state curve was shifted to right by Wenxin granule in the concentration of 10 g/L, which also prolonged the recovery time from inactivation.Conclusions Wenxin granule can block ICa-L, which may Contribute to its antiarrhythmic effect.
