羊痘病毒PCR检测方法的建立和试剂盒的研制

Construction of PCR for Detection of Capripox Virus and its Kit

根据GenBank上已发表的羊痘病毒(CPV)的全基因序列,设计并合成了一对能特异性扩增羊痘病毒的引物,扩增产物大小约为445bp。通过对PCR方法的优化,建立了检测羊痘病毒的PCR方法,并研制出检测试剂盒。同时对试剂盒的敏感性,特异性和稳定性进行了研究。实验结果表明,该试剂盒具有良好的敏感性和特异性,稳定性在-20℃至少可以保存一年。

On the basis of published genomic sequence of capripox virus in the GenBank, a pair of primers for specific amplification of capripox virus fragment were designed and PCR assay for detection of the virus was constructed. PCR amplified product was 445bp. And,the test kit to detect capripox virus was developed. The experiments about kit＇s sensitivity, specificity and expiration date were conducted. The result suggested the kit had high sensitivity and specificity. It could be stored at least for one year at -20℃.
