石刁柏辐射诱变的遗传学分析与RAPD研究 被引量:2

摘要 以60Co-γ作为辐射源,对石刁柏愈伤组织进行5个不同剂量的辐射诱变,经细胞学鉴定和RAPD分析,不同辐射剂量的辐射诱变效果有明显差异。采用细胞学鉴定染色体结构变异,辐射剂量为1、5、10、20 Gy的材料变异率分别为9.4%、36.5%、31.2%、48.2%。在石刁柏辐射品种的RAPD鉴定中,从100个随机引物中筛选出32个有效引物,共扩增出184条DNA片断,其中25条具有遗传多态性,利用Statistica软件计算出遗传离距为0.912~0.983,平均距离为0.952,遗传距离较近,可能与其来自同一个品种的辐射有关。遗传距离结果显示不同辐射剂量石刁柏材料的遗传距离与辐射剂量成一定的正比关系,与细胞学鉴定结果相符。 The callus of Asparagus was radiated by 5 differnt does of 60Co-γ rays form 1Gy to 20 Gy,and made different effect.Corresponding to 1Gy,5Gy,10Gy,20Gy radiation does,the variation of chromosome up to 9.4%,36.5%,31.2%,48.2%.The study identified the different does callus by RAPD.The study selected 32 availability primers from 100 random primers,and produced 184 DNA bands.With statistica software made the result that 25 of them of polymorphism and the similarity coefficient of them were 0.912~0.983.The average similarity coefficient of them is 0.952.It was previously that the genetic resemblance was low as they were sharing common ancestry.The genetic distance showed that the different characters of asparagus in proportion to the radiation doses.The result was in line with cytology expertise.
