摘要
以金铁锁幼茎为外植体进行根茎发生诱导,筛选金铁锁无菌苗诱导、增殖和生根培养的最佳培养基。结果表明:外植体诱导的最佳养基为B5+6-BA0.5 mg/L+IAA0.5 mg/L;分生芽继代增殖的最佳培养基为B5+6-BA0.5 mg/L+IAA0.5 mg/L+Kt0.2 mg/L;最佳生根培养基为B5+NAA0.1 mg/L和IBA0.05 mg/L,生根率83%;最佳培养条件为24℃和12 h/d光照时间。
Optimization of culture media for the induction,multiplication,and rooting of P.tunicoide seedling was conducted using young shoots of P.tunicoide as explants.The results showed that the formula of B5+6-BA0.5 mg/L+IAA0.5 mg/L was suitable for the induction of shooting of P.tunicoides from explants,and B5+6-BA0.5 mg/L+IAA0.5 mg/L+Kt0.2 mg/L for the multiplication of shootings,B5+NAA0.1 mg/L+IBA0.05 mg/L for rooting of P.tunicoides,and in addition,the rooting rate was more than83%.It is proved to be better when the above culture were under a 12 h photoperiod of 2 000 Lx at 24 ℃.
