慢性心力衰竭心肌纤维化大鼠模型的建立 被引量:11

ESTABLISHMENT OF MYOCARDIAL FIBROSIS RATS MODEL WITH CHRONIC HEART FAILURE

摘要 目的:建立成熟、稳定的慢性心衰心肌纤维化大鼠模型。方法:采用肾上腹主动脉缩窄法制作雄性Wistar大鼠慢性心衰模型,随机分为心衰1d组、7d组和14d组,另设立假手术组作为对照。观察各组大鼠左心室肌重量指数(LVMI)和血流动力学参数,HE染色和Masson染色观察左室心肌形态结构的变化。结果:心衰各组大鼠血流动力学参数(LVEDP、±dp/dtmax)与假手术组比较差异显著(P<0.01),并且随着心衰时间的延长,大鼠心功能逐渐恶化;HE染色和Masson染色显示,心衰各组大鼠左室心肌出现明显的病理变化,心肌胶原容积分数(CVF)与假手术组比较有显著差异(P<0.01),且随着心衰时间的延长CVF逐渐升高;心衰各组大鼠LVMI明显高于假手术组(P<0.01),且随着心衰时间的延长逐渐升高。结论:肾上腹主动脉缩窄法建立的慢性心衰心肌纤维化大鼠模型与人心肌纤维化的病理过程相似,可用于心肌纤维化发病机制及药物治疗的研究。 Objective: To establish mature and stable model of myocardial fibrosis with chronic heart failure in rats. Methods:The chronic heart failure Wistar rats model was established by partially banding of abdominal aorta superior to renal artery, then the mrs were randomly divided into 3 groups: heart failure 1d, 7d and 14d group; at the same time, sham-operation group was taken as control. The left ventricular mass indexes (LVMI) and hemodynarnic parameters were observed; HE staining and Masson staining were used to observe the morphological changes of myocardium of left ventricle.Results:There had significant differences between each model group and sham group in hemodynamie parmneters(LVEDE ±dp/dtmax) (P〈 0.01); with time of heat tailure, the heart function of heart failure model rats gradually got worse. There appeared obvious pathological changes of myocardium of rats in model group by HE and Masson staining. There had significant differences between each model group and sham group in collagen volume fraction(CVF )(P〈 0.01); moreover, the CVF gradually increased with time of heart failure. LVMI of rats in each model group was obviously higher than that in sham operation group (P〈0.01), and it gradually increased with time of heart failure.Conclusions:The model of myocardial fibrosis with chronic heart failure in rats established by partially banding of abdominal aorta is similar with the pathological process of human myocardial fibrosis, and can be used to study the pathogenesis of myocardial fibrosis and its medication.
