银杏叶提取物对大鼠胶质瘤作用机制的体内实验研究 被引量:4

摘要 目的探讨银杏叶提取物对大鼠脑胶质瘤疗效及其抗肿瘤机制,为银杏叶提取物的临床应用提供依据。方法培养大鼠C6胶质瘤细胞及建立SD大鼠脑胶质瘤模型;将荷瘤鼠随机分为3组,腹腔内分别注射银杏叶提取物、顺铂及二甲基亚砜(对照组),观察各组大鼠行为改变,记录生存期;头MRI扫描对比注药前后肿瘤体积变化,计算银杏叶提取物组和顺铂组肿瘤抑制率;免疫组织化学方法检测胶质瘤中增殖细胞核抗原(proliferating cellnuclearantigen,PCNA)和血管内皮生长因子(vascular endothelial growth factor,VEGF)的表达。结果成功建立了胶质瘤模型,对照组大鼠行为显著异常,3组生存期银杏叶提取物组>顺铂组>对照组,差异有统计学意义(P<0.05);顺铂组和银杏叶提取物抑瘤率分别达36.2%和43.7%,与对照组相比,差异有统计学意义(P<0.05);经银杏叶提取物和顺铂治疗的大鼠肿瘤中的PCNA和VEGF表达低于对照组,差异有统计学意义(P<0.05,P<0.01)。结论银杏叶提取物能抑制大鼠颅内C6胶质瘤细胞生长,作用机制可能与抑制胶质瘤细胞增殖及诱导肿瘤细胞凋亡有关。 Objective To explore the therapeutic effect and its anticancer mechanism of the ginkgo bioba extract on the C6 brain glioma of rats, and to provide an evidence for its clinical application. Methods C6 glioma cells were cultured and the model of brain glioma in the SD rats was established. The tumor- bearing rats were divided into Ginkgo biloba extract group, Cisplatin group and control group. Ginkgo biloba extract, cisplatin and dimethysulfonide were given by intraperitoneal injection respectively. Their conduct was observed and survival time was recorded. The tumor volume change was observed after administration and tumor control rate was calculated. The expression of proliferating cell nuclear antigen (PCNA) and vascular endothelial growth factor (VEGF) were detected in the glioma by immunohistochemistry. Results The brain glioma model in the SD rats was established. Significant behavior abnormality was found in the control group compared with the treatment groups. The survival time and tumor control rate of the treatment groups were better than those of the control group (P 〈 0. 05 ). The expression of PCNA and VEGF were sisnificantly decreased in the treatment groups ( P 〈 0. 05 ). Conclusion The ginkgo biloba extract can suppress the C6 brain glioma in rats through of inhibition tumor cell proliferation and induction apoptosis.
