联合应用前列腺素E1和还原型谷胱甘肽对大鼠肺缺血再灌注损伤的保护作用 被引量:2

Effects of the Combined Therapy of Reduced Glutathione and Prostaglandin E1 on Long Ischemia-reperfusion Injury in the Rats

摘要 目的观察联合应用前列腺素E1和还原型谷胱甘肽对大鼠肺缺血再灌注损伤的保护作用。方法 90只雄性清洁级SD大鼠随机分为5组,假手术组(S),缺血再灌注组(IR),治疗组1(RG1),治疗组2(RG2),治疗组3(RG3)。每组分别于夹闭缺血的第45分钟、缺血再灌注后1h、2h 3个时间点处死大鼠,取10%左肺匀浆测定肿瘤坏死因子(TNF-a)、肺组织干/湿重比值(D/W)及在光镜下观察肺组织病理变化。结果 (1)TNF-α含量在IR组缺血再灌注后含量明显增加,较S、RG组显著增高(P<0.05);(2)IR组肺组织损伤进行性加重,毛细血管充血、肺泡间隔炎性细胞浸润、肺泡腔内炎性细胞及炎性液体渗出显著,RG组肺组织充血水肿及炎性细胞浸润较IR组减轻,RG3组较RG1以及RG2组减轻;(3)IR组和RG各组的肺组织D/W比值呈进行性下降(P<0.05)。结论联合应用前列腺素E1和GSH对大鼠肺缺血再灌注损伤的保护作用优于单独使用。 Objective To study the protective effects of the combined therapy of reduced glutathione and prostaglandin E1 on long ischemia-reperfusion injury in the rats. Methods Ninety SPF SD rats were divided into five groups randomly,sham operation group (S),ischemia-reperfusion group (IR)and reduced glutathione group (RG1),reduced prostaglandin E1 (RG2),reduced glutathione and prostaglandin E1 group (RG3). Six rats were put to death at one of three time points (45 min ischemia, land 2 hour reperfusion) in each group and the left lung was taken. The tumor necrosis factor alpha(TNF-α ),the dry/wet (D/W) ratio and lung biopsies were al- so obtained. Results (1)The TNF-α in lung tissue:in IR group TNF-α was increased obviously than in the S and RG group(P〈 0.05). (2)Histological evaluation in the process of ischemia-reperfusion the lung injury was aggravated progressively in the IR group; there was marked pulmonary capillary congestion,interstitial edema and intra-alveolar hemorrhage. RG3 was better than RG1 and RG2. (3)After ischemia-reperfusion,the D/W in the IR group and RG group were decreased progressively(P〈0.05). Conclusion Preservation of allograft rats lung with combination of glutathione and prostaglandin E1 is better than single administration for protect lung ischemia-reperfusion injury.
