Effect of Agaricus blazeiMurill Polysaccharides on Expression of Perilipin in Rats With Type 2 Diabetes Mellitus

摘要 目的:探讨姬松茸多糖(ABPS)对2型糖尿病(2-DM)大鼠胰岛素抵抗和脂滴包被蛋白(Perilipin)基因表达的影响。方法:将Wistar大鼠随机分为5组,正常对照组(A组)、模型组(B组)、ABPS高剂量组(C组)、ABPS中剂量组(D组)、ABPS低剂量组(E组)。8周后,测定FBG、FINS和血脂水平;RT-PCR检测Perilipin基因的表达水平。结果:与模型组比较,ABPS组FBG、FINS、TC、TG、LDL-C、FFA降低,Perilipin基因的表达增加(P<0.05)。结论:ABPS可改善糖脂代谢紊乱,降低胰岛素抵抗,其作用可能是通过提高Perilipin基因表达实现。 Objective:To explore the effect of Agaricus blazeiMurill polysaccharides(ABPS)on insulin resistance(IR)and the expression of Perilipin gene in rats with type 2 diabetes mellitus.Methods:Wistar rats were randomly divided into normal control group(group A),diabetic model group(group B),ABPS high dose group(group C),ABPS median dose group(group D)and ABPS low dose group(group E).Levels of fasing blood glucose(FBG),fasing insulin(FINS)and blood lipid were measured after 8 weeks,and expression of Perilipin gene was determined by RT-PCR.Results:Compared with those of model control group,levels of FBG,FINS,TC,TG,LDL-C and FFA in ABPS groups decreased,the mRNA expression of Perilipin increased(P0.05).Conclusions:ABPS might improve carbohydrate and lipid metabolism disorder and decline IR probably through up-regulating the expression of Perilipin gene.
