基于线粒体DNA控制区(mtDNA D-loop)序列分析瓢鸡的遗传多样性 被引量:33

摘要 采用PCR产物直接测序技术对云南省镇沅县50只瓢鸡样品的线粒体DNA控制区(mtDNA D-loop)第Ⅰ高变区序列进行了分析。结果表明:在所分析的D-loop区序列中(520 bp),T,C,A,G平均含量分别为30.4%,29.6%,27.4%,12.6%;共检测到27个核苷酸多态位点,其中24个为转换,3个为颠换,序列突变率为5.19%;序列存在13种单倍型,单倍型多样度(H)为0.883±0.028,核苷酸多样度(π值)为0.015 03±0.000 99,表明瓢鸡群体内遗传变异较丰富。瓢鸡样品可划分为A,B,E,F,G等5个世系,在NJ无根系统树上聚为5大枝,揭示瓢鸡在遗传组成上具有5个母系血统来源。 In the present paper,the PCR products of the mitochondrial D-loop sequences(520 bp in length) of 50 individuals from Piao chicken(which have no pygostyle) of Zhenyuan County,Yunnan province,China,were directly sequenced.The results revealed that the average percentage values of base composition of T,C,A and G in the sequences were 30.4%,29.6%,27.4% and 12.6%,respectively.In the samples assayed,27 mutation sites included 24 transitions and 3 transversions viz.5.19% mutation rate and 13 haplotypes were found,haplotype diversity(H) was 0.883±0.028 and nucleotide diversity(π value) was 0.015 03±0.000 99.Phylogenetic analysis by using the former study and the unrooted NJ method showed that all the sequences were clustered in 5 major clades.The results in this study indicated that the genetic variability in Piao chicken population was high and there were 5 maternal origins to this chicken.
