种子消毒方法和激素处理对和田大叶紫花苜蓿植株再生的影响 被引量:6

摘要 研究了不同种子灭菌方法及次氯酸钠消毒时间对种子萌发率、污染率的影响,并取经灭菌后萌发15-20 d的无菌苗叶片作为外植体,比较了不同激素处理对和田大叶紫花苜蓿愈伤组织诱导及生长的影响。结果表明:(1)流水下冲洗1 h,70%的酒精处理50 s,10%的次氯酸钠震荡22 min是最佳的种子灭菌方法。(2)初步建立和田大叶紫花苜蓿的再生体系如下:诱导愈伤培养基为MS+2,4-D 2.0mg/L+KT 0.5 mg/L+琼脂0.8%;诱导芽分化培养基为MS+6-BA 0.5 mg/L+NAA 0.05 mg/L+琼脂0.9%;生根培养基为1/2MS+IBA 1 mg/L+蔗糖1.5%+琼脂0.8%;出愈率可达到100%;分化率可达到28.3%。 The effect of different sterilization methods,disinfection time and hormone treatments on seed germination and plant regeneration were studied by using the Aseptic seedlings leaf as explants.The results showed that the best method for seed sterilization was rinse materia under running water for an hour with 70% alcohol within 50 seconds,then shaking with 10% sodium hypochlorite in 22 minutes.The regeneration system of Hetian alfalfa as follows:Induction callus culture medium of MS+2,4-D 2.0 mg/L+KT 0.5 mg/L+agar 0.8%;The shoots were induced in the medium of MS+6-BA 0.5 mg/L+NAA 0.05 mg/L+agar 0.9%;The rooting medium of 1/2 MS+IBA 1 mg/L+sucrose 1.5%+agar 0.8%.The Callus rate reached 100% and the differentiation rate reached to 28.3%.
