摘要
建立一种合成和厚朴酚的方法:以4-溴-苯甲醚与4-烯丙基苯甲醚为原料;4-溴-苯甲醚在2,5-二甲基苯基溴化镁、异丙基氯化镁、40℃条件下进行反应,然后加入烯丙溴,得到4-溴-2-烯丙基苯甲醚;4-烯丙基苯甲醚在一定条件下溴代生成2-溴-4-烯丙基苯甲醚;将4-溴-2-烯丙基苯甲醚制成相应的格氏试剂,在NiCl2(PPh3)2催化下与2-溴-4-烯丙基苯甲醚进行交叉偶联,得到和厚朴醇甲醚,水解和厚朴醇甲醚即得到和厚朴酚。
This article provides one way for the synthesis of honokiol: taking 4-Bromo Anisole and 4-allylanisole as raw materials,4-Bromo Anisole had chemical reaction under 40 ℃ in THF and isopropyl magnesium chloride.Then added with allyl-bromide,4-bromo-2-allyl anisole was obtained.Under certain conditions,4-allylanisole generated 2-bromo-4-allyl anisole.Formulating 4-bromo-2-allyl anisole to corresponding Grignard reagent so as to cross-coupling with 2-bromo-4-allyl anisole after been catalyzed by NiCl2(PPh3)2,Honokiol methyl ether was obtained thusly.After hydrolyzing Honokiol methyl ether,honokiol was generated.
