肝硬变患者TGF-β_1基因在外周血中表达及与PⅢP血清水平的关系 被引量:1

摘要 目的:为验证肝纤维化动物模型和慢性肝病患者肝组织中,转化生长因子(TGF-β1)的mRNA表达增强。作者分析肝硬变患者外周血TGF-β1mRNA和血清中有活性的TGF-β1蛋白及其结果与型前胶原N端肽(PP)血清水平间的关系。方法:肝硬变患者50例,健康自愿者8人,取其外周血,分离单核细胞(PBMC),提取总RNA,用巢式RT-PCR扩增TGF-β1mRNA并测定血清中TGF-β1和PP的水平。结果:分析表明患者外周血单核细胞(PBMC)中TGF-β1mRNA有高阳性率的表达。血清中TGF-β1与PP的水平均明显高于对照。结论:研究结果证实肝硬变患者TGF-β1基因在外周血中表达增强,与PP血清水平相关,表明TGF-β1可能在肝纤维化。 Objective:The aims of the present study were to determine the expression of transformation growth factor β 1 (TGF β 1) mRNA and TGF β 1 protein in the peripheral blood and its relation to serum levels of procollagen type Ⅲ N terminal peptide (PⅢP) in the patients with cirrhosis. Methods:Peripheral blood samples were obtained from 50 patients with cirrhosis and 8 normal healthy volunteers. Total RNA was extracted from PBMC and TGF β 1 mRNA was amplified from total RNA by nested RT PCR. Serum levels of TGF β 1 and PⅢP were determined. Results:TGF β 1 mRNA could be detected in the peripheral blood mononuclear (PBMC) with high positive rate and serum levels of TGF β 1 and PⅢP were significantly higher than controls. Conclusion:These results confirm the increased expression of TGF β 1 and the positive correlation between TGF β 1 and PⅢP in the patients with cirrhosis. And our results support the suggestion that TGF β 1 may be a significant factor in the increased collagen systhesis of human fibrosis.
