反相高效液相色谱法同时测定红松种仁中的三种内源激素 被引量:1

Simultaneous Determination of Three Endogenous Hormones in Pinus koraiensis Nuts by Reversed phase High Performance Liquid Chromatography

摘要 建立了红松种仁中赤霉素、吲哚乙酸、脱落酸等3种内源激素的反相高效液相色谱检测方法。采用HiQ-Sil C_(18)柱(4.6 mm×250mm,5μm),以甲醇-水-冰乙酸(45.0:54.2:0.8,V:V:V)混合溶液为流动相,流速为1.0mL/min,进样量为10μL,检测波长为254 nm,柱温为40℃,用外标法进行定量测定。GA_3、IAA和ABA的最低检测限分别为0.5 mg/L、0.2 mg/L和0.1 mg/L,平均回收率分别为95.2%、97.2%和94.1%,相对标准偏差分别为2.3%、1.7%和1.3%。该方法具有简便、准确和易操作等特点。 A reversed-phase high performance liquid chromatography (RP-HPLC) method was developed for the simultaneous determination of gibberellin acid (GA3), indole-3-acetic acid (IAA), and abscisic acid (ABA) in Pinus Koraiensis nuts. The method was performed with a HiQ-SiL C18 column (4.6mm×250mm,5μm) using methanol- water-acetic acid (45.0 : 54.2 : 0.8, V:V:V) as mobile phase, sample injection at the velocity of 1.0 mL/min with an input of 10 μL, and detection wavelength at 254 nm. ,column temperatuve 40℃.The detection limit of GA3, IAA and ABA were 0.5, 0.2 and 0.1mg/1, respectively. The average recoveries were 95.2 %, 97.2 % and 94.1%, respectively. The relative standard deviations were 2.3 %~ 1.7 % and 1.3 %, respectively. The method is convenient, fast and accurate.
