摘要 褪黑激素、复合胺及吲哚乙酸等神经递质在高等植物中的发现为理解植物生理的机制提供了新的可能性。建立了一种在线制备方法,将样品粉末化后装填入小柱,利用水、甲酸-水和甲醇-水在线预洗制备后,切换至分析流路,采用梯度洗脱及ESI源LC-MS/MS检测植物中3种神经递质,每个样品的检测时间10 min,较离线方法的分析时间缩短了1/2,且不损失方法的准确度及精密度;回收率较离线方法提高约4%。 The discovery of plant neurotransmitters,melatonin,serotonin,and indole-3-acetic acid in higher plants has given a new possibility in understanding the physiology of plants.In this study,we report an on-line method for extraction of melatonin,serotonin and indole-3-acetic acid in plant.Sample was eluted by gradient and quantified by LC-MS/MS with ESI source after grounded plant packed in column,as well as pilot online cleanout and preparation with water,acidic water,and methanol solution step by step.The total run time for each sample was 10 min,while shortened half time compared with off-line method without losing accuracy and precision of method.The recovery of on-line method can be enhanced about 4% more than off-line.All three compounds were quantified with on-line sample preparation,and a reliable result was achieved for routine application.
