柴红丸治疗黄褐斑小鼠模型的实验研究

Experimental Study of Chaihong Pill in the Treatment of Chloasma Mouse Model

目的观察柴红丸对实验性黄褐斑模型小鼠的肝脏和皮肤中超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量的影响。方法将60只SPF级NIH雌性小鼠,按体重随机分为空白组、模型组、百消丹组以及柴红丸高、中、低剂量组,每组10只。除空白组动物外,其余动物采用肌肉注射黄体酮加紫外线照射法致黄褐斑实验动物模型。动物分组,分别按剂量灌胃给药30 d后,处死动物并分别取肝脏和背部皮肤各一块,检测各组小鼠肝脏和皮肤中SOD活性和MDA含量。结果柴红丸高、中剂量组能明显升高黄褐斑模型动物的肝脏和皮肤中SOD活性(P<0.01)、降低MDA含量(P<0.01);柴红丸低剂量能明显升高黄褐斑模型动物皮肤中SOD活性(P<0.01)、降低肝脏和皮肤中MDA含量(P<0.01)。结论柴红丸对实验性黄褐斑模型小鼠有较好的治疗作用,其机制可能与升高抗氧化酶活性、降低过氧化物含量有关。

Objective To observe the impacts of chaihong pill on superoxide dismutase（SOD）activity and malondialdehyde（MDA）content in the liver and skin of chloasma mouse model.Methods 60 NIH female mice of SPF grade were randomized into blank group,model group,baixiaodan group and chaihong pill groups of high,medium and low doses,10 mice in each one according to the body weight of mice.Except blank group,intramuscular injection of progesterone and ultraviolet irradiation were adopted in all of the rest animals to induce chloasma model.After group division,the animals were sacrificed after gastric lavage administration with different doses for 30 days.Afterwards,the liver and a piece of skin on the back were collected for the determination of SOD activity and MDA content in the liver and skin in each group.Results Chaihong pill high-dose group and medium-dose group could increase obviously SOD activity（P0.01）and reduce MDA content（P0.01）in the liver and skin of chloasma mouse model.The low-dose group could increase apparently SOD activity in the skin and reduce MDA content of the liver and skin（P0.01）of chloasma mouse model.Conclusion Chaihong pill presents the good efficacy on experimental chloasma mouse model probably through increasing SOD activity and reducing MDA content.
