小青龙汤和人参五味子汤二步序贯治疗对哮喘小鼠呼吸道炎症与重塑的影响 被引量:13

Effects of Xiaoqinglong Decoction and Ginseng-Schisandrae Decoction by Two-Step Sequential Therapy on Airway Inflammation and Remolding in Mice with Asthma

摘要 目的探讨小青龙汤和人参五味子汤二步序治疗贯治疗对哮喘小鼠呼吸道炎症与重塑的影响。方法健康雌性昆明系小鼠40只,随机分为4组(每组10只):正常对照组、哮喘组、布地奈德组和二步序贯治疗组。采用卵清蛋白建立小鼠哮喘模型;在致敏阶段,二步序贯治疗组小鼠胃饲人参五味子汤;在激发阶段,布地奈德组小鼠在激发前雾化吸入布地奈德,二步序贯治疗组予以小青龙汤胃饲。采用普通光镜观察哮喘小鼠呼吸道组织病理变化确定呼吸道炎症;免疫组织化学法测定基质金属蛋白酶-9(MMP-9)、基质金属蛋白组织酶抑制剂-1(TIMP-1)水平,确定呼吸道重塑程度。结果正常对照组小鼠呼吸道组织颜色呈粉红色,其余各组小鼠呼吸道组织较正常对照组略苍白,以哮喘组较明显,部分出现黑色斑点或纹理,体积增大。布地奈德组和二步序贯治疗组呼吸道组织炎症改变均较哮喘组明显减轻,差异有统计学意义(Pa<0.01);但二组较正常对照组明显加重,差异有统计学意义(Pa<0.01);布地奈德组与二步序贯治疗组呼吸道炎症改变比较差异无统计学意义(P>0.05)。布地奈德组和二步序贯治疗组呼吸道MMP-9、TIMP-1表达水平和MMP-9/TIMP-1明显低于哮喘组,差异有统计学意义(Pa<0.01);但二组与正常对照组比较,呼吸道MMP-9和MMP-9/TIMP-1明显增高,差异均有统计学意义(Pa<0.01);布地奈德组与二步序贯治疗组各指标差异均无统计学意义(Pa>0.05)。结论二步序贯治疗对哮喘小鼠具有抑制呼吸道炎症和重塑的作用,在一定程度上可以替代布地奈德吸入治疗。 Objective To explore the effects of Xiaoqinglong decoction and Ginseng-Schisandrae decoction by two-step sequential therapy on airway inflammation and remolding in mice with asthma.Methods Forty healthy female Kunming mice were divided into 4 groups randomly(n=10):normal control group,asthmatic group,budesonide group and two-step sequential therapy group.The asthmatic model were established by ovalbumin;Ginseng-Schisandrag decoction was administered to mice in two-step sequential therapy group by gastogavage in sensitization stage;in stimulation stage,budesonide atomization was administered to mice in budesonide group,and Xiaoqinglong decoction was administered to mice in two-step sequential therapy group by gastrogavage before stimulation.Then the changes of pathology in airway tissue were observed by using ordinary microscope to confirm inflammation.The levels of matrix metalloproteinase-9(MMP-9) and tissue inhibitor of metalloproteinase-1(TIMP-1) of airway tissue were detected by immunohistochemical method to confirm airway remodelling.Results The airway tissues of the mice in normal control group were pink,while the color in other groups was slightly pale,in which asthmatic group was relatively obviously pale,with black spots or texture in some airway tissues of the mice in asthmatic group,and the volume of some airway tissues in asthmatic group increased to varying degrees.Pathological method of the lung showed that the inflammatory cell infiltration in treatment groups were less than that in asthmatic group(Pa0.01),but more than that in normal control group(P0.01).There was no significant difference between the budesonide group and two-step sequential therapy group(P0.05).The MMP-9,TIMP-1 and MMP-9/TIMP-1 were decreased in the budesonide group and two-step sequential therapy group compared with those in asthmatic group(Pa0.01),and the MMP-9 and MMP-9/ITIMP-1 were increased compared with those in the normal control group(Pa0.01).There was no significant difference between budesonide group and two-step sequential therapy group(Pa0.05).Conclusion Xiaoqinglong decoction and Ginseng-Schisandrae decoction by two-step sequential therapy may restrain airway inflammation and remolding,which can replace budesonide to a certain extent.
