摘要
目的研究吗啡条件性位置偏爱期间疏肝解郁方对大鼠前额叶皮质和伏核中谷氨酸受体1(GluR1)、谷氨酸受体2(GluR2)蛋白表达的影响。方法采用Western blot实验方法定量检测GluR1和GluR2蛋白表达的变化。结果与正常对照组比较,吗啡模型组GluR1和GluR2的蛋白表达均明显升高(P<0.01)。与吗啡模型组比较,疏肝解郁方对吗啡模型组GluR1和GluR2的蛋白表达均明显下调,差异具有统计学意义。结论吗啡模型组大鼠前额叶皮质和伏核GluR1和GluR2的蛋白表达增多,参与了吗啡依赖的形成过程。而疏肝解郁方对前额叶皮质和伏核GluR1和GluR2的蛋白表达具有明显下调作用,干预了吗啡依赖的形成。
Objective To study the influences of Shuganjieyu Formula on the protein expressions of glutamate receptor 1(GluR1) and glutamate receptor 2(GluR2) in the prefrontal cortex and nucleus accumbens in rats during morphine conditioned place preference period.Method The protein expression changes of GluR1 and GluR2 were detected quantitatively by applying Western blot method.Result The protein expressions of GluR1 and GluR2 increased significantly in the morphine group compared with the control group(P0.01).The protein expressions of GluR1 and GluR2 decreased significantly in the Shuganjieyu Formula group compared with the morphine group and the difference was significant statistically.Conclusion The protein expressions of GluR1 and GluR2 in the prefrontal cortex and nucleus accumbens increased in the rats of the morphine group,which participated the formation of morphine dependence.Shuganjieyu Formula has a significant down-regulation effect on the protein expressions of GluR1 and GluR2 and intervenes the formation of morphine dependence.
