摘要
制备黄曲霉毒素M1污染样品的净化前处理免疫亲和柱。利用无血清培养基制备的高纯度抗黄曲霉毒素M1单克隆抗体和溴化氰活化的Sepharose 4B进行偶联、装柱后通过IC-ELSIA对免疫亲和柱的性能评价确立了最佳反应条件。免疫亲和柱的有效柱容量为500 ng,经添加回收试验测定的平均回收率为92.46%,变异系数2.3%~7.2%。利用高纯度抗黄曲霉毒素M1单克隆抗体和溴化氰活化的Sepharose 4B制备的免疫亲和柱可应用于乳制品等食品中黄曲霉毒素M1污染度测定的样品快速前处理。
Preparation of aflatoxin M1 immunoaffinity column for clean-up contaminated samples.The high-purity anti-aflatoxin M1 monoclonal antibody with serum-free medium was prepared and coupled with CNBr activated Sepharose 4B,and then packed in a column.Then the column was evaluated through IC-ELISA and established the optimum reaction conditions.The capacity of immunoaffinity column was 500 ng,the average recovery of aflatoxin M1 was 92.46%;and the variation coefficients were 2.3%~7.2%.The immunoaffinity columns was prepared by coupling high-purity anti-aflatoxin M1 monoclonal antibody with CNBr Activated Sepharose 4B can be applied to detect contamination samples fast,such as aflatoxin M1 in dairy products.
