摘要
以从海洋中分离筛选得到的细菌Flavobacteriaceae sp.CZ1127为研究对象,通过单因素实验和正交试验对其产海参岩藻聚糖硫酸酯酶的培养基成分进行优化,提高其产酶量。得出其最佳培养基组成为(w/v):海参岩藻聚糖硫酸酯0.25%、酵母粉0.15%、MgSO4.7H2O 0.05%、CaCl2 0.02%、KCl 0.01%、Na2HPO4 0.22%、NaH2PO4 0.06%、NaCl 2.5%;确定了该菌株的最佳培养条件为:500 mL的三角瓶装液150 mL、温度为25℃、转速150 r/min、发酵时间为48 h。优化后,海参岩藻聚糖硫酸酯酶活力为22.88 U/mL,比优化前提高了3.01倍。
Flavobacteriaceae sp.CZ1127,a fucoidanase-producing strain,was isolated and the optimal conditions for fucoidanase fermentation by Flavobacteriaceae sp.CZ1127 were optimized by single-factor experiments and orthogonal experiments.The optimum culture medium was obtained as: sea cucumber fucoidan 0.25%,yeast 0.15%,MgSO4.7H2O 0.05%,CaCl2 0.02%,KCl 0.01,Na2HPO4 0.22%,NaH2PO4 0.06%,NaCl 2.5%.After cultivated in 150mL medium with 500mL shake flask at 25 ℃for 48 h with shaking speed of 150 r/min,the activity reached the highest value of 22.88 U/mL which was 3.01 times higher than that under the original conditions.
