解毒益智胶囊的益智效应及对中枢胆碱能神经功能的影响 被引量:3

The Intelligence Promotion Effect and the Influence on Central Cholinergic Neuronal Function of Jieduyizhi Capsules

摘要 目的观察解毒益智胶囊的益智效应及对中枢胆碱能神经功能的影响,为治疗轻度认知障碍提供实验依据。方法建立东莨菪碱致小鼠学习记忆障碍模型。按体重随机分为正常对照组、模型对照组、安理申对照组及解毒益智胶囊高、中、低、极低剂量组。采用避暗法、跳台法和Morris水迷宫法观察解毒益智胶囊的益智效应;采用乙酰胆碱酯酶(AchE)试剂盒检测全脑AchE活性;采用实时荧光定量PCR技术,以18S rRNA为内参基因,用SYBR GreenⅠ染料法检测海马M1受体和胆碱乙酰转移酶(ChAT)基因相对表达,双标准曲线法进行计算和数据分析。结果解毒益智胶囊能显著延长东莨菪碱致模型小鼠学习记忆障碍模型避暗实验和跳台实验逃避潜伏期,减少错误次数,显著缩短Morris水迷宫实验平均逃避潜伏期,改善空间搜索策略;能显著提高学习记忆障碍小鼠模型全脑AchE的活性;显著增强海马M1受体和ChAT基因表达,解毒益智胶囊高、中、低、极低剂量组M1受体基因分别较模型组表达增高了14.9、7.3、8.2、1.8倍,而ChAT基因则分别较模型对照组提高了6.6、26.5、1.2、6.1倍。结论解毒益智胶囊具有良好的益智作用。 Objective To explore the intelligence promotion effect and the influence on central cholinergic neuronal function of Jieduyizhi (JDYZ) Capsule in order to provide experimental basis for its treatment on mild cognitive impairment (MCI). Method The mice model with learning and memory deficits induced by scopolamine was established. The step through test, step down test and the Morris water maze test were applied on mice to assay the intelligence promotion effect of JDYZ Capsule. The AchE activity in the whole mice brain was tested by Ache ldts. The relative gene expression of ChAT and M1 receptor in the hippocampus was tested by FQ-PCR (Time Fluorescent Quantitative PCR) and SYBR Green I method, with the 18sRNA as a reference gene and analyzed by double-standard curve method. Result JDYZ Capsule can effectively prolong the escape response latency in the mice model with learning and memory deficits induced by scopolamine, reduce the number of errors, significantly reduce the average escape response latency in the Morris water maze test, and improve the strategies on searching the surrounding space. JDYZ Capsule remarkably improved the Ache activity of mice brain in the learning and memory deficit model. The gene expression of ChAT and M1 receptor in the hippocampus were improved. Compared with the control group, the level of M1 receptor gene expression in the high-dose, middle-dose, low-dose and extreme low-dose were separately increased by 14.9 times, 7.3 times, 8.2 times and 1.8 times, while the level of ChAT gene expression were increased by 6.6 times, 26.5 times, 1.2 times and 6.1 times. Conclusion JDYZ Capsule has favorable effect on intelligence promotion.
