摘要
对在非水相中经固定化碱性蛋白酶Alcalase 2.4L水解的玉米蛋白肽的体外醒酒活性进行了研究。考察了在体外试验中,玉米肽对乙醇脱氢酶(ADH)及对乙醛脱氢酶(ALDH)活力的影响,分析了活性最高的酶解物的氨基酸组成和相对分子量分布及其与醒酒活性之间的关系。结果表明:当控制水解度最高为22.7%时,非水相中制备的玉米肽在体外试验中对乙醇脱氢酶(ADH)和乙醛脱氢酶(ALDH)的激活作用最高,ADH激活率可达27.1%,ALDH激活率可达50.0%,此高醒酒活性肽中含有较高比例的Ala、Leu及Pro,尤其是Leu,3种氨基酸含量均高于相同水解度下水相中制备的玉米肽的氨基酸含量。在酶解产物分子量<3 548 u,其中分子量<1 295 u的肽含量最多,约占80%,以2~10肽为主。
Crude corn peptides were prepared via immobilized enzymatic hydrolysis of corn proteins in nonaqueous media,and its alcohol dehydrogenase and acetaldehyde dehydrogenase activities in vitro were studied.Amino acids composition and relative molecular weight distribution of the peptides were analyzed.Results showed that the highest alcohol dehydrogenase and acetaldehyde dehydrogenase activation ratio were 27.1% and 50.0%,respectively,when the hydrolysis degree reached 22.7%.Corn peptides contained high proportion of alanine,praline and leucine,especially leucine.Molecular weight of enzymatic hydrolysis products were less than 3 548 u,the most were below 1295 Da,taking up about 80%,mainly be 2~10 peptides.
