丙戊酸钠对脊髓小脑性共济失调3型细胞模型的神经保护 被引量:3

摘要 目的探讨在脊髓小脑性共济失调3型(spinocerebellar ataxia 3,SCA 3)可诱导细胞模型中,丙戊酸钠(valproic acid,VPA)的神经保护作用。方法构建可诱导性SCA 3细胞模型PC12Tet-On-Ataxin-3-Q 28/84,MTT法比较细胞存活率。ImageJ软件测定细胞轴突长度(n=100),Western blot检测Acetyl histon H 3的表达水平。结果 Q 84组随Dox诱导时间延长存活率降低(P<0.05),而Q 28组无明显差异(P>0.05)。Q 84组诱导病理性ataxia-3表达细胞存活率下降,VPA处理后可提高细胞存活(P<0.05)。Q 84组Acetyl histon H 3的表达受抑制,加入VPA后,Acetyl histon H 3表达量明显增多(P<0.05)。结论 VPA可提高SCA3可诱导细胞模型的细胞存活率,促进轴突再生,VPA作为组蛋白去乙酰化抑制剂可提高Acetyl histon H 3的表达。
