摘要
目的:观察人绒毛膜癌(绒癌)细胞系中BeWo细胞株在福斯考林(forskolin)诱导融合前后细胞中基质金属蛋白酶-2(MMP-2)及上皮钙黏蛋白(E-cad)的表达变化,以探讨绒癌细胞融合与其浸润和转移能力变化的关系。方法:用BeWo细胞株进行培养传代,以100mmol/L浓度的forskolin作用于BeWo细胞,用逆转录聚合酶链(RT-PCR)方法检测细胞中人类内源性逆转录病毒基因编码的膜糖蛋白(Syncytin mRNA)表达量的变化;免疫印迹技术(Western blot)方法检测细胞中表达MMP-2及E-cad蛋白表达量的变化。结果:以BeWo细胞作为靶细胞,根据RT-PCR和Western blot方法中条带的形状和亮暗程度可知Syncytin基因和E-cad及MMP-2蛋白的表达呈时间依赖性,随着forskolin作用时间的增加,Syncytin基因及E-cad蛋白的表达逐渐增加,而MMP-2蛋白表达量逐渐减少,差异具有统计学意义(P<0.05)。结论:Syncytin高表达与融合后绒癌细胞的浸润转移能力的改变可能有一定关系,Syncytin的检测有望成为滋养细胞肿瘤患者判定预后的有价值的指标,并为滋养细胞肿瘤的治疗开辟了新的思路。
Objective:To observe the changes of the expressions of MMP-2 and E-cad before and after Forskolin treating the Human Choriocarcinoma BeWo cell line,to investigate the relationship of the fusion status of Choriocarcinoma cells and their invasive and metastatic capabiligy. Methods:BeWo cell lines were cultured and passaged,then were treated by 100mmol/L Forskolin. Syncytin mRNA was detected by RT-PCR,also the changes of the protein,MMP-2 and E-cad were checked by Western blot. Results:According to the shape and brightness of PCR and Western bands,Syncytin mRNA and E-cad,MMP-2 protein showed time-dependent changes. Along with the reaction of Forskolin,the expression of Syncytin mRNA and E-cad protein increased steadily,while the expression of MMP-2 protein decreased,the differences were significant (P〈0.05).Conclusions:The high expression of syncytin might be related with the changes of invasive or metastatic capability post fusion of choriocarcinoma cells. So Syncytin might be a valueble prognostic marker and offer a new treatment ragimen for trophoblastic tumor.
