摘要
以卷曲霉素(CPM)产生菌卷曲链霉菌S.capreolus4006(效价为6640u/mL)为出发菌,经UV诱变/五氯酚浓缩,获得一株效价为7950u/mL的营养缺陷型菌株406-35,再经EMS诱变,筛选到一株抗5000u/mLCPM、效价为8513u/mL的突变株5-41,经原生质体NTG诱变处理,获得一株效价为9050u/mL的高产菌N13菌株N13连续传代10代,卷曲霉素效价保持稳定通过单因子试验和多因子正交试验,确定了菌株N13的最佳发酵条件菌株N13在最佳发酵条件下卷曲霉素效价为9483u/mL,比出发菌S.
Using capreomycin producing Streptomyces capreolus 4 006 (with the yield of capreomycin being 6640 u/mL) as parent strain, after UV mutation and eutrophication with pentachlorophenol, an auxotrophic strain 4006-35 producing 7 950 u/mL of capreomycin was obtained. Subsequently the auxotrophic strain 4006-35 was treated with EMS, and the mutant 5-41 resistant to 50 000 u/mL of CPM was obtained and the mutant produced 8 513 u/mL of CPM. After the protoplasts of strain 5-41 were treated with NTG, a mutant strain N13 producing 9 050 u/mL of capreomycin was obtained and strain N13 was stable in capreomycin production after continuous subcultures for 10 generations. Optimum fermentation conditions were established based on the study on fermentation conditions for strain N13. Under the optimum conditions, the capreomycin yield by N13 was 9483 u/mL and increased by 42.8% compared with the original parent strain 4006 under the original fermentation conditions.
