摘要
目的探讨CD3+、CD8+、CD20+细胞在二乙基亚硝胺(DEN)造模大鼠肝硬化、肝癌、肝癌肺转移3个阶段肝组织中的数量及意义。方法 DEN制备肝硬化、肝癌、肝癌肺转移大鼠模型,免疫组织化学(SP)法进行CD3、CD8、CD20染色,对阳性细胞进行定量分析。结果 CD3+细胞平均数从高到低为:肝癌组织、肝癌肺转移组织、肝硬化组织、正常肝组织(P<0.05);CD8+细胞平均数从高到低为:正常肝组织、肝硬化组织、肝癌组织,肝癌肺转移(P<0.05);CD20+细胞平均数从高到低为:肝癌组织、肝癌肺转移、肝硬化组织、正常肝组织(P<0.05)。结论在DEN制备的大鼠模型中,随着肝组织进一步恶化,CD3+、CD20+细胞逐渐增多,CD8+细胞却减少,导致肿瘤微环境免疫状态的紊乱。
Objective To investigate the significance of CD3+,CD8+,CD20+ cells in DEN made mould rat of liver cirrhosis,liver cancer,liver cancer metastasis three stages.Methods CD3+,CD8+,CD20+ cells were detected in DEN model of rats by immunohistochemical staining with streptavidin-horseradish peroxidase detec-tive system.Results The order of the number of CD3 positive cells was in the tissues of live cancer,lung metastasis of hepatocellular carcinoma,liver cirrhosis,normal liver(P0.05).The order of the number of CD8 positive cells was in the tissues of normal liver,liver cirrhosis,live cancer,lung metastasis of hepatocellular carcinoma(P0.05).The order of the number of CD20 positive cells was in the tissues of live cancer,lung metastasis of hepatocellular carcinoma,liver cirrhosis,normal liver(P0.05).Conclusions In the DEN made rat models of liver cirrhosis,with the worsening of the liver cancer,the number of CD3 and CD20 positive cells elevated gradually,but declined for the CD8,which lead the disorder of the immune state of the microenvironment of liver cancer.
