Expression of the matrix metalloproteinases 7 and 9 in the prostate cancer

摘要 目的探讨基质金属蛋白酶(MMP)-7,-9在前列腺癌转移侵袭中的作用,以及血管内皮生长因子(VEGF)与MMPs的关系。方法采用免疫组织化学和形态定量分析方法分析MMP-7及MMP-9表达情况,同时培养PC3M细胞并采用Boyden小室侵袭试验及酶谱分析观察MMP-9及其与VEGF在前列腺癌中的作用。结果前列腺癌各分级中MMP-7和MMP-9的表达差异均有统计学意义(P<0.01)。Boyden小室侵袭实验显示,不同试验组穿透Matrigel的细胞数依次递减,即PC3M>PC3M+MMP-9Ab(1∶100)>PC3M+MMP-9Ab(1∶50),差异均有统计学意义(P<001)。酶谱分析观察发现,直接培养于培养皿表面的PC3M细胞仅有稳定的MMP-9和MMP-2酶原表达,但PC3M细胞与VEGF共孵育组条件培养基中MMP9酶原含量以VEGF剂量依赖的方式增加。结论 MMP-7和MMP-9可作为肿瘤侵袭和转移的判定指标。VEGF与MMPs的分泌有关,其机制有待于进一步探讨。 Objective To investigate the expression of the matrix metalloproteinases(MMP)-7 and-9 in the pro-state cancer,and the relationship of between the MMP and vascular endothelial growth factor(VEGF).Methods The expression of the MMP 7 and 9 were detected by immunohistochemistry assay and morphology quantitative analysis technique.At the same time,the PC3 M cells were cultured,the expression of MMP-9 and the relationship between MMP-9 and VEGF were detected with Boyden chamber and zymography assay.Results The expression of the MMP-7 and MMP-9 were different in each graduation of prostate cancer(P0.01).The result of the Boyden chamber showed that the number of the penetration Matrigel decreased gradually in different group [PC3MPC3M+MMP-9Ab(1∶100)PC3M+ MMP-9Ab(1∶50)].The result of zymography assay showed that the expression of MMP-9 and MMP-2 were stable in the PC3M cells cultured in the culture dishes,but in the VEGF co-cultured culture dishes,the content of the MMP-9 was elevated dose-dependently.Conclusion MMP7 and MMP-9 can be used as the indicators of prostate cancer invasion and metastasis,and the VEDF was related with the secretion of the MMPs.
