摘要 [目的]观察HEK293细胞系中颗粒蛋白前体(PGRN)对核因子-κB(NF-κB)的活性影响.[方法]在HEK293细胞中采用瞬时过表达PGRN、瞬时的通过RNA干扰来下调PGRN、采用报告基因系统共同来研究PGRN在影响NF-κB活性过程中所行使的具体功能.[结果]HEK293细胞中本底状态能检测到PGRN的表达,过表达PGRN不会增加NF-κB本底状态下的活性,而通过RNA干扰来对内源性PGRN进行下调后,则发现NF-κB活性显著的降低.[结论]PGRN可能在基线水平参与了NF-κB的活性调节. [Objective] To observe the effect of progranulin(PGRN) on nuclear factor kappa B(NF-κB) activity in HEK293 cell lines, [Methods] Transient overexpression of PGRN and transient siRNA for down regulating PGRN as well as reporter gene system were conjointly used to investigate the specific function of PGRN in the progress of PGRN affecting NF-κB activity in HEK293 cells. [Results] The basal level of PGRN expression in HEK 293 cells could be detected. The overexpression of PGRN did not increase the activity of NF-κB at baseline. However, the ac tivity of NF-κB significantly decreased after siRNA was used to down regulate the endogenous PGRN. [Conclusion] PGRN may be involved in the regulation of the activity of NF-κB at baseline.
