摘要 目的应用甲基化特异性PCR(MSP)方法检测非小细胞肺癌(NSCLC)CDH13基因启动子甲基化情况。方法留取44例非小细胞肺癌患者手术标本及12例非肺癌患者正常肺组织,MSP分析CDH13基因启动子甲基化情况。结果 44例非小细胞肺癌中CDH13基因启动子甲基化阳性率为54.5%(24/44),12例正常肺组织中未检测到此基因的甲基化(0/12)。CDH13基因甲基化与患者性别及吸烟与否无关;肿瘤分期越晚甲基化的发生率越高。结论原发性非小细胞肺癌中存在着较高比例的CDH13启动子甲基化,提示CDH13在非小细胞肺癌的发生中起着重要作用。 Objective To detect the methylation status of CDH13 promoter region in non-small cell lung cancer by using methylation-specific PCR technique(MSP).Methods The tissue specimens from 44 cases of lung cancer,and 12 cases of normal tissues were examined,and CpG island methylation of CDH13 gene was detected by methylation specific PCR.Results Methylation of CDH13 promoter was detected in 44(54.5%) cancer samples,the incidence of promoter hypermethylation was not found in 12 benign lung disease tissues.Anymore,hypermethylation of CDH13 was not associated with gender and smoking.The incidence ratio of promoter hypermethylation of CDH13 was ascendant with the cancer becoming much later.Conclusion Frequency of CDH13 promoter methylation is apparently higher in lung cancer tissues than that in normal tissues.It signifies that CDH13 methylation may contribute to development of lung cancer.
