摘要
【目的】研究证实肌腱损伤修复过程中病灶区病理学变化及BFGF和Ⅰ型胶原蛋白在肌腱损伤修复过程中的作用。【方法】构建Leghorn鸡屈趾肌腱损伤修复模型,借助体视学、生物力学以及免疫组化进行探察和评价,实验数据利用SPSS10.0软件统计处理。【结果】①肌腱粘连程度和抗张力强度分别随损伤修复时间的延长逐渐增加,第6周组明显高于其它时间组(P<0.001),而且各时间组均与对照组之间存在显著统计学差异(P<0.001);②组织病理学检测显示病灶区出现大量炎症细胞和肌腱细胞,除第6周组外,其它时间组肌腱细胞和巨噬细胞的数量均高于对照组(P<0.05),同时显示肌腱细胞数量的峰值出现于第2周(P<0.001),而巨噬细胞数量在各时间组之间无统计学差异(P>0.05)。③免疫组化探测显示BFGF阳性细胞数量及Ⅰ型胶原蛋白的表达在各时间组均高于对照组(P<0.05);而且在第2周组显著高于其它时间组(P<0.001)。【结论】肌腱损伤修复过程中肌腱粘连程度和抗张力强度的变化与损伤修复时间正相关,病灶区组织病理学变化、BFGF和Ⅰ型胶原蛋白表达的高峰出现于第2周,提示BFGF和Ⅰ型胶原蛋白牵涉到损伤肌腱的修复过程。
【Objective】 In order to confirm the pathological changes in the lesion areas,and to investigate the function of BFGF and collagen type Ⅰ in the process of tendon injury and repair,the present study constructed the Leghorn chicken model of tendon injury and repair and carried out the exploration and evaluation utilizing the stereology,biomechanics,and immunohistochemistry.【Methods】All the experimental data were statistically analyzed with SPSS10.0 software.【Results】Our results showed that the degree of tendon adhesion and tensile strength gradually increased as the process of injury and repair continuing,and these degrees of the 6th week group were apparently higher than that of other experimental groups(P 0.001).Compared them between different time groups and the control group P 0.001.Large inflammatory and tendon cells were observed in the lesion areas,and the number of tendon cells in the 1-4 weeks groups were higher than that in the control group(P 0.05).The number of tendon cells reached the peak at the end of 2nd week(P 0.001),but the variance of macrophages quantities was not statistically significant among different time groups(P 0.05).The number of BFGF positive cells and expression of collagen type I were higher in all experimental groups than that in the control group(P 0.05),and these results of the 2 weeks group were obviously higher than that of other experimental groups(P 0.001).Our results indicated that in the process of tendon injury and repair,the degree of tendon adhesion and tensile strength were positively correlated with the repair time.Histopathological changes and expression of BFGF and collagen type I reached the peak at the end of 2nd week.【Conclusion】These characteristics suggested that BFGF and collagen type I was correlated with the repair of injured tendon.
