数字化珊瑚羟基磷灰石人工骨的研制及体外细胞毒性研究 被引量:6

Preparation and Cytotoxicity Evaluation of Digital Coral Hydroxyapatite Artificial Bone in Vitro

摘要 目的研究数字化珊瑚羟基磷灰石(CHA)人工骨材料的制备方法,并评价其细胞毒性。方法将珊瑚羟基磷灰石微粒与左旋-聚乳酸通过不同的质量比混合均匀后,采用选择性激光烧结的工艺,制备数字化CHA人工骨材料。采用体外细胞培养技术,观察其浸提液对L-929细胞形态的变化,采用MTS法检测对L-929细胞增殖情况的影响,以评价其细胞毒性。结果采用选择性激光烧结快速成型的方法可以制备出不同质量比的数字化CHA人工骨,其浸提液对体外培养的L-929细胞形态无明显影响,对细胞生长和增殖无抑制作用,其细胞毒性均为0级。结论采用选择性激光烧结快速成型的工艺可以制备出不同质量比的数字化CHA人工骨,且该材料没有细胞毒性,具有作为可吸收人工骨材料的应用潜力。 Objective To investigate the preparative method of the digital coral hydroxyapatite artificial bone and evaluate its cytotoxicity in vitro, in order to provide a sound scientific basis for clinical use. Methods The micropartical of coral hydroxyapatite and L-Polylactide were well mixed, and the digital coral hydroxyapatite artificial bone was prepared by sintered the mixture, using elective laser sintering rapid prototyping (SIS RP) process. L929 cells were cultured with the extracts of the digital coral hydroxyapatite artificial bone with different masses in vitro. The morphologic changes of L929 cells were observed by inverted microscope. MTS tests were performed to evaluate the cell proliferation, and the relative growth rate (RGR) was calculated to evaluate the cytotoxicity of the digital coral hydroxyapatite artificial bone with different masses.Results The digital coral hydroxyapafite artificial bone with different masses could be prepared by elective laser sintering rapid prototyping process. No side effects were found in growth, proliferation and cell morphology of L929 cells cultured with the extracts of the digital coral hydroxyapatite artificial bone with different masses in vitro. The cytotoxicity tests showed that cytotoxi- city graduation of the digital coral hydroxyapatite artificial bone with different masses was 0 grade.Conclusion The digital coral hydroxyapatite artificial bone with different masses can be prepared by elective laser sintering rapid prototyping process, and the artificial bone has no cytotoxicity. It is a prospective artificial bone.
