摘要
目的:建立银翘柴桂汤剂中黄芩苷、绿原酸、芍药苷的含量测定方法。方法:采用HPLC法,色谱柱为Kromasil Cl8(4.6 mm×250 mm,5μm);流动相为甲醇-1%磷酸水(梯度洗脱);流速1 mL·min-1;柱温为35℃;多波长检测芍药苷检测波长230 nm,黄芩苷检测波长280 nm,绿原酸检测波长327 nm。结果:绿原酸、芍药苷和黄芩苷在40 min内均能与其他组分实现良好分离,绿原酸、芍药苷和黄芩苷分别在0.077 6~0.776 0μg(r=0.999 9),0.076 1~0.761 0μg(r=0.999 9),0.264 8~2.648μg(r=0.999 9)线性关系良好,加样回收率分别为102.67%(RSD 1.48,n=6),97.85%(RSD 1.85,n=6),101.86%(RSD1.44,n=6)。结论:方法简便、结果准确,能同时测定银翘柴桂汤中绿原酸、芍药苷和黄芩苷3种有效成分的含量。
Objective:To develop an HPLC method for the Content determination of chlorogenic acid,paeoniflorin and baicalin inYinqiaochaigui decoction.Method:HPLC-based separation of the agents was performed on Kromasil Cl8 column(4.6 mm × 250 mm,5 μm) at 25 ℃ with the mobile phase of methanol-0.1% Phosphoric acid water solution(gradient elution),flow rate of 1.0 mL·min^-1 and multiple detection wavelength of 327,230,280 nm.Result:HPLC allowed simultaneous quantitative determination of the 3 components in Yinqiao Chaigui decoction,and they showed good linear relationships when their sample amount ranged 0.077 6-0.776 0 μg,0.076 1-0.761 0 μg and 0.264 8-2.648 μg,respectively,with correlation coefficients all beyond 0.999 9 and average recovery rates of 102.67%,97.85% and 101.86%,respectively.Conclusion:This method is simple and convenient in detecting the 3 components in Yinqiao Chaigui decoction,and the results are precise and reproducible.
