HIV-1/AIDS患者外周血T细胞CD127分子的表达及其临床意义

Expression of CD127 of T lymphocytes in peripheral blood of patients with HIV-1/AIDS and its clinical significance

目的观察CD127分子在未治疗和经高效抗逆转录病毒治疗(HAART)的HIV-1/AIDS患者外周血CD4+、CD8+T细胞上的表达水平,并探讨其临床意义。方法采用流式细胞术检测48例未治疗的和57例经HAART治疗的HIV-1/AIDS患者外周血CD4+和CD8+T细胞表面CD127分子的表达,以36名健康正常人作对照;同时用荧光定量PCR技术检测HIV-1/AIDS患者血浆中HIV-1病毒载量。结果未经治疗组其T细胞CD4+ CD127+和CD8+ CD127+的表达明显低于治疗组和对照组,且与外周血HIV-1病毒的载量相关,病毒载量越高其CD127分子的表达水平越低;HAART治疗后,CD127在CD4+和CD8+T细胞上的表达水平明显上升,显著高于未治疗组,但仍低于对照组的水平;HAART治疗后,CD127分子在T细胞表面的表达水平与CD4+T细胞数成正相关,外周血CD4+细胞数越多,其T细胞表面的CD127的表达率越高。结论 HIV/AIDS患者外周血CD127分子表达明显减少,HAART治疗后能有效地促进CD127分子在T细胞上的表达,一定程度上能重建T细胞亚群功能及数量。

OBJECTIVE To study the expression of CD127 on CD4^＋ and CD8^＋ T lymphocytes of HIV-1/AIDS patients with or without HAART,and the relationship with disease progression.METHODS A cross-section study of 48 AIDS patients without HAART and 57 AIDS patients receiving HAART was carried out and healthy people were recruited as controls.The percentage of CD4^＋CD127^＋ and CD8^＋CD127^＋ T cells was tested using flow cytometry.HIV-RNA in plasma of HIV-1/AIDS patients was detected by quantitative RT-PCR.RESULTS The percentage of CD4^＋CD127^＋ and CD8^＋CD127^＋ T cells in AIDS patients without HAART was higher than that of AIDS patients receiving HAART and controls,which decreased with the increase of HIV-1 load.The percentage of CD4^＋CD127^＋ and CD8^＋CD127^＋ T cells in AIDS patients receiving HAART was still lower than controls.Expression of CD127 on T lymphocytes was related to CD4^＋T cell count,which was highly expressed with higher CD4^＋T cell count.CONCLUSION Expression of CD127 on peripheral lymphocytes of HIV-1/AIDS patients decreases significantly and can be increased by effective HAART,which can stimulate the count and function of T lymphocytes of HIV-1/AIDS patients.
