摘要
目的观察"益气调血,扶本培元"针法对多发性梗塞痴呆(MID)模型认知功能的改善作用。方法采用栓子注入法制作MID模型,将雄性Wistar大鼠随机分为正常组、假手术组、模型组、针刺组和非穴组,每组15只。针刺组选取膻中、中脘、气海及双侧血海、足三里;非穴组选取双侧肋下各2个固定非穴点,每日1次,治疗6天,休息1天,共治疗3周。采用Morris水迷宫检测造模后大鼠的学习记忆功能;免疫组化法观察各组大鼠海马CA1区、CA3区、DG区及大脑皮层葡萄糖转运蛋白1(GLUT1)的表达。结果模型组大鼠学习记忆能力显著下降(P<0.05);而与模型组与非穴组比较,针刺组大鼠发现平台所需时间明显缩短(P<0.05或P<0.01),跨越原平台次数和原平台区域的停留时间均明显增加(P<0.05)。模型组大鼠各脑区GLUT1的表达量明显高于正常组和假手术组(P<0.05);除皮层外,针刺组大鼠海马CA1、CA3、DG区的GLUT1表达量与模型组及非穴组比较差异均有统计学意义(P<0.05)。结论 "益气调血,扶本培元"针法能够上调GLUT1表达,促进葡萄糖的跨膜转运,从而增强血管性痴呆大鼠脑组织的葡萄糖代谢,改善脑组织的缺血缺氧,进而改善认知功能。
Objective To observe the cognitive-improving effect of acupuncture for Yiqi Tiaoxue(replenishing qi and regulating blood)and Fuben Peiyuan(reinforcing the anti-pathogenic qi and consolidating the body resistance)on multiple infarction dementia(MID)model rat.Methods The embolic suspension was injected to induce multiple infarct dementia.The male Wistar rats were randomized into the normal group,sham operation group,model group,acupuncture group,and sham acupuncture group,with 15 in each.The acupuncture group was treated with acupuncture at Shanzhong(CV 17),Zhongwan(CV 12),Qihai(CV 6),Xuehai(SP 10),and Zusanli(ST 36).The sham acupuncture group was treated with acupuncture at the two non-acupoints in the bilateral hypochondrium.The treatment was once a day for 6 days in succession and 1 day for rest as interval,3 weeks in total.The learning and memory function of model rats was tested with Morris water maze.The expression of glucose transporter 1(GLUT-1)in the cortex and hippocampus CA1,CA3,and DG areas was detected by immunohistochemical method.Results The learning and memory ability of MID rats was obviously impaired(P0.05).Compared with the model group and sham acupuncture group,the acupuncture group spent less time in finding the platform(P0.05 or P0.051),the number of jumping through the original platform and the staying time on the original platform were significantly increased(P0.05).The expression of GLUT-1 in the model group was significantly increased than that in the normal group and sham operation group(P0.05).Except in the cortex,the expression of GLUT-1 in the hippocampus CA1,CA3,and DG areas of the acupuncture group was significantly different from that of the model group and sham acupuncture group(P0.05).Conclusion The acupuncture for Yiqi Tiaoxue and Fuben Peiyuan is effective to improve the cognitive function of MID rat through up-regulating the expression of GLUT-1 for promoting the transmembrane transport of glucose so as to enhance the glucose metabolism and relieve the ischemia and hypoxia of the brain tissues of vascular dementia rat.
