摘要
旨在分析水分胁迫对小麦叶片抗病相关酶的诱导,探讨水分胁迫诱导抗病性的机制。以水分胁迫可诱导其抗病性的小麦品种-小偃6号为材料,用不同质量分数PEG-6000模拟干旱胁迫,采用真空渗透离心法分离胞间隙液以及胞内蛋白,并对不同胁迫时间胞间隙和胞内抗病相关蛋白及酶活性进行分析。结果显示,随着胁迫强度的增加和处理时间的延长,叶片及胞间隙液POD、PPO、PAL(胞间隙除外)、几丁质酶活性均表现出明显的上升的趋势,复水2 d后下降,但各项测定指标均未恢复至对照水平。由此可知,水分胁迫可诱导小偃6号叶片中各种抗病防卫相关酶活性的增强,表明小麦对水分胁迫的响应与抗病信号转导途经之间可能存在共同的机制。
In order to analyse enzymes related with disease resistance in wheat leaves induced by drought,and explore the mechanisms of drought induced disease resistance.Wheat variety Xiaoyan 6 with the character of drought induced stripe rust resistance was treated with different concentrations of PEG-6000 to simulate drought stress.The intercellular washing fluids and intracellular proteins were isolated using vaccum penetration-centrifuge method.Disease-related proteins and enzymes in intercellular washing fluids and cell were measured at different treatment time.The results showed that with the increase of stress intensity and the prolongation of treatment time,the activities of peroxidase(POD),polyphenoloxidase(PPO),phenylalanine ammonia lyase(PAL) and chitinase in both leaves and intercellular washing fluids(IWF) showed a tendency to increase at first,then decrease after rehydration,however,none index recovered to the level of the control.Meanwhile,drought can induce the activities of various defense-related disease-resistant enzymes in wheat Xiaoyan 6 leaves to enhance its resistance to diseases.There may be common mechanisms underlying the signalling pathway of water stress response and disease resistance.
