摘要
目的研究JAK-STAT、NF-Κβ信号通路在斯氏狸殖吸虫脑病模型大鼠脑组织中的表达及其意义。方法制作SD大鼠肺吸虫脑病模型。将12只SD雄性大鼠随机分为肺吸虫抗原注射组(n=6)、生理盐水对照组(n=6)。用脑立体定位仪微量注射肺吸虫抗原至大鼠大脑皮质建立大鼠肺吸虫脑病模型,对照组注射生理盐水,24 h后解剖大鼠,取脑组织、固定、切片。免疫组织化学方法观察大鼠脑组织中JAK-STAT和NF-Κβ信号通路的表达活性。结果免疫组化显示生理盐水对照组大鼠脑组织中NF-Κβ和JAK-STAT呈弱阳性表达,NF-Κβ和JAK-STAT阳性率分别为4.4%、4.7%;肺吸虫抗原注射组大鼠脑组织中可见STAT和NF-Κβ阳性细胞,细胞染成棕黄色或有棕黄色颗粒沉积,肺吸虫抗原注射组大鼠大脑皮质组织NF-Κβ和JAK-STAT阳性率分别为47.1%和45.8%,以大脑皮质损伤区明显,胞质和胞核中均见阳性表达,抗原注射组和生理盐水组之间差异有统计学意义(χ2=476,χ2=447,P<0.05)。结论肺吸虫在脑组织中机械破坏作用和抗原的致敏作用激活了JAK-STAT、NF-Κβ信号通路的表达,提示STAT的表达和活化在肺吸虫脑病发生过程中可能发挥重要作用。
Objective To study expression and significance of JAK-STAT,NF-Κβ in the brain of experimental rat with Paragonimiasis.Methods Twelve healthy adult male SD rats were randomly divided into model group(n=6) and control group(n=6).Brain damage model group rats were established by injecting Paragonimus adult worm antigen,and control group by injecting saline with stereotactic microinjection apparatus.The rats brain tissue were obtained 24h after,fixed and liced.The expression and activity of the NF-Κβ and JAK-STAT signaling pathway were observed by immunohistochemistry.Results The NF-Κβ and JAK-STAT presented low expression in normal rat brain,and the expressing positive rate of NF-Κβand JAK-STAT was 47.1% and 45.8% respectively.There were visible positive expression cells of STAT and NF-Κβ in the brain of Paragonimus skrjabini antigen injection rat,especially at the cortical damage zone,positive expression could be obseved in the cytoplasm and nucleus,which were stained to yellow brown,and the expressing positive rate of NF-Κβand JAK-STAT in the brain of Paragonimus skrjabini antigen injection rat was 47.1% and 45.8% respectively.There were significant difference between the normal rat and antigen injection rat(X2=476,X2=447,P0.05).Conclusion Paragonimiasis mechanical damage in the brain and antigen sensitization activated JAK-STAT,NF-Κβ expression of signaling pathway,suggesting that STAT expression and activation may play an important role in the progress of Paragonimiasis encephalopathy.
