喉鳞癌caveolin-1和MVD表达及与颈淋巴结转移的关系 被引量:5

Relationship among the expression of the caveolin-1 and MVD with cervical lymph nodes metastasis in laryngeal squamous cell carcinoma

摘要 目的:探讨喉鳞状细胞癌组织中caveolin-1 mRNA及蛋白的表达和微血管密度(MVD)与颈淋巴结转移的关系。方法:采用免疫组化法检测40例喉鳞状细胞癌组织中caveolin-1、CD34表达,并用半定量逆转录聚合酶链式反应(semi-RT-PCR)技术检测以上组织中caveolin-1 mRNA表达水平。结果:不伴颈淋巴结转移的喉癌组(第1组)的caveolin-1 mRNA相对A值为0.450±0.050,伴有颈淋巴结转移的喉癌组(第2组)的caveolin-1 mRNA相对A值为0.701±0.073;caveolin-1蛋白第1组中阳性表达率为25.0%,第2组为65.0%,不论从基因还是蛋白水平,两组相比差异均有统计学意义(均P<0.05)。CD34免疫组织化学染色结果显示第2组的MVD值为30.19±2.60显著高于第1组值21.34±4.62(P<0.05)。喉鳞状细胞癌组织中caveolin-1高表达与MVD升高呈正相关(r=0.875,P<0.05),且二者与颈淋巴结转移相关。结论:caveolin-1在喉鳞状细胞癌中的表达可能在肿瘤转移过程中起了重要作用,与肿瘤血管形成关系密切。 Objective: To study the relation among the expression of the caveolin-1 and microvessel density (MVD) with cervical lymph nodes metastasis in laryngeal squamous cell carcinoma (LSCC). Methods: The expression of eaveolin-1 and CD34 in 40 samples of tumor were detected by immunohistoehemical method. The expression of eaveolin-1 mRNA in samples of tumor was detected by semi-RT-PCR technique. Results: The caveolin-1 mRNA A of LSCC without eervieal lymph nodes metastasis was (the first group) 0.450±0.050, the caveolin-1 mRNA A of LSCC with cervical lymph nodes metastasis was (the second group) 0.701±0.073; the positive rate of the eaveolin-1 protein expression in the first group was 25.0%, in the second group was 65.0%, eaveolin-1 was higher either at molecular level or at the protein level in the second group than the first group (all P〈0.05). The expression of MVD marked by CD34 in the second group was 30.19±2.60, and in the first group was 21.34±4.62, the expression of MVD in the second group was higher than the first group (P〈0.05). The over expression of caveolin-1 had a positive correlation with a higher MVD and were both related to cervical lymph nodes metastasis in the 40 cases (r=0.875, P〈0.05). Conclusion: These results suggest that over expression of caveolin-1 may play a very important role in the metastasis of laryngeal squamous cell carcinoma and is closely correlated with angiogenesis.
