摘要
建立了Fe2+-H2O2-亚甲基蓝化学发光新体系并用于测定保健品对羟自由基的清除率。实验发现,在酸性条件下,Fe2+和H2O2在线生成的羟自由基(.OH)能与亚甲基蓝产生极强的化学发光,加入保健品溶液后可部分清除体系产生的羟自由基,降低化学发光强度。据此,结合流动注射技术,建立了一种简单、快速、高效测定保健品对羟自由基清除率测定的化学发光新方法。对Fe2+-H2O2-亚甲基蓝反应体系进行平行测定(n=11),测得该法的相对标准偏差为0.39%。将该法用于市售保健品清除羟自由基的测评,获得满意结果。
A novel and effective chemiluminescence method for determination of hydroxyl radical scavenging rate was established based on Fe2+-H2O2-methylene blue chemiluminescence system.In an acid media,a strong chemiluminescence signal occurred when methylene blue solution mixed with Fenton reagent,then the signal decreased after the health products was added to the system due to hydroxyl radicals produced from Fenton reagent were scavenged partially in the solution.This phenomenon was the basis of determining hydroxyl radical scavenging rate.The relative standard deviation for 11 measurements of each sample was 0.39%.This method was successfully used to evaluate the antioxidant activity of commercially available health products and good results were obtained.
