摘要
目的比较不同基原吴茱萸温中止痛疗效及其作用机制的差异性和等同性。方法建立水浸拘束法致大鼠应激性胃溃疡模型,通过观察不同基原吴茱萸对大鼠胃溃疡指数、胃酸分泌、血清胃泌素及5-羟色胺的影响,比较并探讨其温中作用的差异及其可能的作用机制。采用冰醋酸诱导的小鼠扭体反应实验和小鼠毛细血管通透性的实验,比较不同基原吴茱萸的镇痛及抗炎作用。结果吴茱萸、石虎和疏毛吴茱萸3种基原吴茱萸水提物2 g.kg-1,对水浸法引起的大鼠胃黏膜损伤均具有保护作用。其中,疏毛吴茱萸在降低溃疡指数,减轻胃酸刺激等方面具有较优效果,其作用机制可能是通过降低血清胃泌素并升高血清5-羟色胺。吴茱萸可以显著降低血清胃泌素。石虎在抗炎止痛方面具有较优的效果。结论 3种基原吴茱萸特征化学成分组成接近,温中止痛的疗效基本一致,为临床应用提供实验依据,但是其作用机制因其主成分间的偏重而有所差异。
OBJECTIVE To compare the efficacy on relieving gastric ulcer and pain among three origins of Euodiae Fructus and to clarify the possible mechanisms.METHODS The water immersion induced irritability gastric ulcer rats model was established.Gastric ulcer index,gastric acid secretion,serum gastrin and 5-HT were observed and compared among three origins of Euodiae Fructus.Acetic acid writhing test and the capillary permeability were tested in mice to compare the anti-inflammatory effects.RESULTS The aqueous extracts of three origins of Euodiae Fructus,i.e Euodia rutaecarpa(Juss) Benth,E.rutaecarpa var.officinalis,E.rutaecarpa var.bodinieri,given at dose of 2 g·kg-1,showed protective effects on gastric mucosa.E.rutaecarpa var.bodinieri showed optimum effect in lowering ulcer index and reducing gastric acid stimulation,whose mechanism may be reduction of serum gastrin and elevation 5-HT.E.rutaecarpa could reduce serum gastrin significantly.E.rutaecarpa var.officinalis showed the optimum effect on relieving inflammatory pain.CONCLUSION The three origins of Euodiae Fructus with similar characteristic components can protect gastric mucosa and relieve pain.The mechanisms may be related to serum gastrin and 5-HT level.
