摘要 目的:建立一种实时在位药物动力学的研究方法—近红外监测系统,并应用常规方法HPLC对其可行性进行验证。方法:利用体外实验验证后的近红外监测系统实时在位监测了大鼠股动脉中cyapte的动力学过程,DAS软件算出药动学参数,并用HPLC方法在相同条件下测得的药代动力学参数作为验证。结果:近红外监测系统实时在位监测了cyapte在大鼠体内的动力学过程,得出的药代动力学参数与HPLC法得出的结果无统计学差异,2种方法具有等效性。结论:近红外监测系统可以应用于药物的实时在位动力学研究,特别是在可用cypate进行标记的蛋白多肽类药物领域具有广阔的应用前景。 Objective:A near-infrared monitoring system for in vivo real time monitoring of the dynamic of drugs was designed,and validated by a routine method—HPLC.Method: The dynamic processes of cypate in rat was in vivo real-time monitored on the femoral artery by the established NIR monitoring system and calculate the pharmacokinetic parameters using the DAS software which validated by the HPLC method.Results: The results of pharmacokinetic parameters of cypate in rat from the two methods have no statistics difference.Conclusion: NIR monitoring system can be applied to in vivo real-time pharmacokinetic study,especially in the field of protein and peptide and give a broad application prospects.
