摘要
目的:建立银杏叶提取物的HPLC-DAD-ELSD多维指纹图谱研究方法。方法:使用SepaxGP-C18(3.0 mm×150 mm,3μm)为分析柱,以1%甲酸水溶液、甲醇和乙腈作为流动相进行梯度洗脱,流速0.4 mL·min-1,柱温35℃;DAD检测器扫描范围为200~400 nm;ELSD检测器氮气流量1.5 L.min-1,漂移管温度60℃,增益值为1。结果:利用该法获得了银杏叶对照提取物的多维指纹图谱,并对5种不同的银杏叶提取物进行了指纹图谱相似度计算,取得满意的效果。结论:该法操作简单、谱图信息丰富、稳定性、重复性好,可作为各类银杏叶原料、中间产物及银杏叶提取物产品的质量控制依据。
Objective: To establish a HPLC-DAD-ELSD method for the determination of fingerprint chromatograms of Ginkgo biloba extracts.Method:A SepaxGP-C18(3.0 mm×150 mm,3 μm)was used as analysis column and 1% formic acid/methanol/acetonitrile as mobile phase at the flow rate of 0.4 mL·min-1 with gradient elution.The column temperature was set to 35 ℃,DAD detector's scan range was between 200 nm and 400 nm,N2 flow rate of ELSD was 1.5 L·min-1,the temperature of drift tube was set to 60 ℃ and the gain number was set to 1.Result:This method was successfully applied to acquire the fingerprint chromatograms of standardized EGb and determine the similarities between standardized EGb and 5 EGbs that from different manufacturers.Conclusion: The method was proved convenient,informative,stable and repeatable.It can be applied to the quality control of ginkgo leaves,its extracts and relative products.
