多指标正交试验优化白首乌C_(21)甾总苷的醋酸水解条件 被引量:2

Optimization of the hydrolysis process for C_(21) steroidal glycoside of Bai Shou Wu by acetic acid with multi-target orthogonal design

摘要 目的:研究白首乌C21甾总苷醋酸水解的最佳条件。方法:以开德苷元-3-O-β-D-洋地黄毒糖苷、告达庭、开德苷元-3-O-α-L-迪吉糖-(1→4)-β-D-磁麻糖苷和告达庭-3-O-β-D-磁麻糖苷的含量为指标,采用正交试验设计L9(33)进行优选,考察水解温度(A)、水解时间(B)、醋酸浓度(C)3个因素对白首乌C21甾总苷醋酸水解条件的影响,同时建立HPLC含量测定的方法。结果:影响醋酸水解的主次因素为A>B>C,最佳水解条件为5.0%醋酸溶液,100℃回流6 h。结论:此水解条件明显提高了白首乌中抗癌有效成分的含量,对研究抗肿瘤药物具有重要作用。 Objective:To study the optimizal hydrolysis process for C21 steroidal glycoside of Bai Shou Wu by acetic acid.Method:The effects of acetic acid concentration,reaction temperature and reaction time had been investigated using orthogonal design and the contents of kidjoranin 3-O-β-digitoxopyranoside,caudatin,kidjoranin 3-O-α-L-diginopyranosyl-(1→4)-β-cymaropyranoside and caudatin 3-O-β-cymaropyranoside as response indexs were determined by the high performance liquid chromatography.Result:The factors influencing acetic extraction efficiency were as follows:ABC(A.reaction temperature;B.reaction time;C.acetic acid concentration).The optimal hydrolysis condition obtained was:refluxing for 6 hours with 5.0% dilute CH3COOH solution at 100 ℃.Conclusion:The content of antitumor active ingredients under the optimum hydrolysis condition is raised obviously and has a great part in studying this antitumor drug.
