摘要
目的:建立RP-HPLC-DAD同时测定太白贝母药材中7种核苷类活性成分(胞苷、尿苷、鸟苷、胸苷、腺苷)及碱基(尿嘧啶、腺嘌呤)含量的方法。方法:采用Zorbax SB-Aq C18色谱柱(4.6 mm×250 mm,5μm),流动相为甲醇和水,梯度洗脱,流速1 mL.min-1,检测波长260 nm,柱温25℃。结果:尿嘧啶、胞苷、尿苷、鸟苷、胸苷、腺苷、腺嘌呤分别在0.312~37.440,1.095~65.700,1.020~61.200,1.075~64.500,0.812~48.720,1.070~64.200,0.432~25.920 mg.L-1与峰面积呈良好线性关系。平均回收率为96.96%~103.5%,RSD≤3.8%。结论:太白贝母有紫外吸收的水溶性成分主要是核苷类成分,所建立方法简单、重复性好,准确可靠,可用于贝母药材中核苷类化合物的含量测定,为全面开发利用贝母药材提供进一步依据。
Objective:To develope a high performance liquid chromatography coupled with a photodiode array detector(HPLC-DAD) method for simultaneous determination of 7 nucleosides and nucleobases in Fritillaria taipaiensis.Method:The analyses were performed on an Agilent Zorbax-SB-Aq-C18 column(4.6 mm× 250 mm,5 μm) eluted with water and methanol in gradient mode.The flow rate was 1.0 mL·min-1.The detection wavelength was 260 nm.The temperature of sample manager was set at 25 ℃,and the injection volume was 20 μL.Result:The investigated compounds including uracil,cytidine,uridine,guanosine,thymidine,adenosine and adenine were shown good linearity(r≥0.999 8) over the tested ranges.The average recoveries were within 96.96%-103.5% with RSD ≤ 3.8%.Conclusion:The accuracy,stability,repeatability and average recovery of the method are satisfying,and the seven nucleosides and nucleobases components in F.taipaiensis can be rapidly and accurately quantified by HPLC-DAD.This work provided helpful information for comprehensive quality evaluation of F.taipaiensis.
